The Donegal Cycle Route starts in Donegal Town connecting to the North West Trail and the National Cycle Network in Donegal Town, and follows a route along the local roads as much as possible through Mountcharles, Inver, Ardara, Dungloe, Loughanure, Crolly, Dunlewy, Creeslough, Glen, Milford, Ramelton, Letterkenny, and connects with the National Cycle Network and Sustrans Route 92 at Newtown Cunningham. There are several signed spurs along the route to nearby towns – Burtonport, Gortahork, and Dunfanaghy and there are also signed loops – The Dooey Way Loop at Leitirmacaward, The Maghery Way Loop at Dungloe and the Bunlin Way Loop at Milford. The Donegal Cycle Route follows Class 2 & 3 county roads as much as possible but there are occasions where the Route has no alternative but to use more major roads. Cyclists are asked to be aware of other road traffic at all times and to obey the rules of the road, on all classes of road used.
